My top ten (from the perspective of someone born and raised in Philly and living near DC eliminating KC, StL and Denver which are high on my list): Boston in early autumn, Seattle/Portland/Vancouver whenever it's relatively dry, San Diego in January (beach+city+Tijuana), Miami/FL Keys, Albequerque/Santa Fe, Arizona, Yellowstone, Dallas, San Francisco, New York in Spring. For the record, DC is a wonderful pace to visit during the Cherry Blossom Festival in early spring.

My wife and I went on a long weekend trip to Seattle. I'd recommend it. You can take the light rail from the airport to downtown where there is plenty to do. Lunch or Dinner on the Space Needle, EMP museum, Pikes fish market, etc...and everything is within walking distance. They also have a monorail that takes you from downtown to the Space Needle/museum area.
You'd love the EMP museum. They have a really cool Sci-Fi area there.
