I don't know if they will use The Punisher in The Defenders. Krysten Ritter is saying that Jessica Jones season two and The Defenders will be filmed very closely together though:

Speaking to Digital Spy, Krysten Ritter says that the Netflix crossover show and the second season of Jessica Jones will shoot back to back:

They’ll be back-to-back. I don’t know if they’ll overlap a little bit. It’s about to be a serious undertaking. I’m starting my physical training - lots of weight training and boxing.

Meanwhile, she also teased the possibilities the second season could bring—including appearances from other Marvel characters:

I’m excited to see what happens next for Jessica, and the end of season one left a lot of doors open and questions unanswered. Kilgrave was her reason for getting up every morning.
